Gene Therapy (2026)
In combination with our VTX-PID, we have generated new insights into anti-AAV3B neutralizing antibodies to expand eligibility for AAV gene therapy :
1- AAV3b shows strong tropism for human hepatocytes, but pre-existing neutralizing antibodies (NAbs) remain a key barrier to patient eligibility.
2- Our new study maps both seroprevalence and NAb titer distribution in healthy adults and Wilson’s disease patients. In healthy adults, 33–36% were NAb-positive, and 19% of seropositive individuals had intermediate titers (1:5–1:45). This intermediate-titer group may be convertible to eligible patients through IgG-cleaving enzyme–based NAb-reduction approaches. In Wilson’s disease, only 17.4% of patients were NAb-positive.
3- These data help define target-population eligibility and support strategies to broaden access to systemic AAV3b-based gene therapies and other AAVs with similar cross reactivity. Further, these results indicate a clear strategy to expand patient eligibility by including those with intermediate NAb titers and transiently reducing their NAbs with VTX-PID to enable effective AAV transduction.
4- These findings are critical for clinical trial design and for accurately estimating the treatable patients for all systemic AAV gene therapies.
